Lorenzo led every lap at the Spanish circuit while Rossi was forced to accept third place following a frantic battle with Dani Pedrosa throughout the race.
Lorenzo claimed the lead from pole man Marc Márquez and surged into the lead, though his Honda rival quickly clawed back the deficit and emerged as a threat.
However, Márquez lost the front end of his bike at Turn 12 and crashed out, reacting furiously to his own error.
That left Lorenzo clear up front, a few seconds ahead of Pedrosa and Rossi, who quickly got by fast-starting Andrea Iannone.
Rossi made his first move on Pedrosa with five laps remaining into Turn 5 but the Spaniard fought back, a situation which was repeated a few laps later.
Having already tried a couple of times at Turn 1, Rossi made a block pass into Turn 5 to move into second, but Pedrosa dived up the inside at Turn 7 and edged Rossi onto the kerbs.
Rossi tried once more further around the lap but ran too deep, allowing Pedrosa to ease back through into second place and return to the podium places.
